Publications on PNAS and Nature Protocols : development of a novel method called NAD tagSeq to identify NAD-capped RNAs
NAD is a coenzyme and plays a central role in regulating cellular redox balance. In the past decade, it has been found that some of the 5' end of RNAs was capped with NAD. The NAD cap could mediate RNA activity and impact biological functions.
 
In 2019, the SKLEBA members, Prof. Yiji Xia and Prof. Zongwei Cai worked together to develop a method called NAD tagSeq to identify the NAD-capped RNAs in Arabidopsis thaliana. This work has been published on PNAS (https://doi.org/10.1073/pnas.1903683116). The paper is the first work to integrate the Oxford Nanopore direct RNA sequencing method to identify the NAD-capped RNAs. It also demonstrates that most of the NAD-capped RNAs in A. thaliana are protein-coding RNA.
 
Recently, Prof Zongwei Cai, Prof. Yiji Xia, Mr. Shao Xiaojian, Mr. Hailei Zhang submitted the detailed experimental design and procedure of NAD tagSeq to Nature Protocols. The manuscript has been accepted last month and now in the press.
